Commute from Sienna to Downtown or Medical Center

Just wanted to ask someone living in Sienna (or knows someone that is currently) how the commute is from Sienna to downtown or medical center during rush hour (lets say you want be there at 8 AM and leave around 5-5:30PM). I know the maps app can be unreliable with their estimates vs real world experience. Also what is the toll prices each way (I am guessing it will be FB tollway to 288 for the fastest commute). Lastly, I am guessing there will be a big difference being in the front of Sienna vs back of Sienna.
